BCG vaccinatie studie – help ons de respons op vaccinatie beter te begrijpen in de strijd tegen
tuberculose!
Tuberculose komt toch niet meer voor. Waarom dit onderzoek?
Tuberculose komt helaas nog wel veelvuldig voor, niet heel veel in Nederland, maar wel daarbuiten. De
infectie komt ook steeds dichterbij, niet alleen door reizen en migratie, maar ook door een toename in
moeilijk te behandelen gevallen, bijvoorbeeld door resistentie voor antibiotica.
Dagelijks overlijden meer dan 4000 mensen aan tuberculose en dat is veel te veel. In de strijd tegen
tuberculose en andere infectieziekten willen we beter begrijpen hoe het immuunsysteem reageert voor
en na vaccinatie met het BCG vaccin. BCG wordt al meer dan 100 jaar gebruikt, maar nog steeds hebben
we vragen over de reactie op dit vaccin.
De kennis die we in het BCG vaccinatie onderzoek zullen opdoen is niet specifiek voor tuberculose, maar
zal daarnaast ook toegepast kunnen worden in het verbeteren van vaccins tegen andere infectieziekten.
Een win-win situatie.
Het BCG vaccinatie onderzoek
Het BCG vaccinatie onderzoek wordt uitgevoerd in het LUMC.
We willen gezonde vrijwilligers (18-55 jaar, mannen en vrouwen) vaccineren met BCG. We weten dat
mensen heel verschillend kunnen reageren op de BCG vaccinatie, maar weten niet waarom die reacties
zo verschillend zijn.
Wat gebeurt er?
Alle deelnemers worden kosteloos gevaccineerd met:

Enkele dosis rabiës vaccin na 6 maanden
Voor de BCG vaccinatie wordt een klein huidbiopt afgenomen, bij alle bezoeken zal bloed worden
afgenomen, en een uitstrijk van de mondholte gemaakte worden (swab).
In de huid zullen we kijken welke cellen van het afweersysteem aanwezig zijn en of die verschillen van
persoon tot persoon. In het bloed zullen we de functie van het afweersysteem onderzoeken, zowel voor
als na vaccinatie.
Hoeveel tijd kost het?
Als je meedoet dan duurt het BCG-vaccinatie onderzoek voor jou 7,5 maand en daarin vraagt het
studieteam je om zeven keer langs te komen bij het LUMC voor de vaccinatie en studiebezoeken.
Het eerste studiebezoek duurt ongeveer een half uur, het 2 e bezoek een uur en de overige bezoeken
ongeveer een kwartier. Er is een onkostenvergoeding beschikbaar.
Wanneer kan je meedoen?
Je kan meedoen als:

Je tussen de 18 en 55 jaar bent,

Je in de omgeving van Leiden woont
-
Je gezond bent
-
Je niet eerder gevaccineerd bent met BCG of tegen rabies
Ook ben je bereid om gedurende 7,5 maanden 7 keer langs te komen.
Welk vaccin wordt gebruikt?
De vaccins die gebruikt worden heten BCG (Danish strain 1331) en Rabipur (tegen rabiës, Bavarian
Nordic A/S).
Beide vaccins zullen in de huid (intradermaal) worden toegediend. Voor BCG is dit de standaard manier,
voor Rabipur is dat een methode die niet de standaard is maar waarmee wel heel veel ervaring is in het
LUMC. Rabipur wordt al meer dan 30 jaar gebruikt en is bewezen veilig en effectief.
Let op: De rabiësvaccinatie die u krijgt in dit onderzoek biedt geen volledige bescherming tegen rabiës,
omdat slechts een enkele dosis gegeven wordt. Wilt u bescherming tegen rabiës, neem dan contact op
met de GGD.

BCG Vaccination Study – help us better understand the response to vaccination in the fight against
tuberculosis!
Tuberculosis doesn’t occur anymore, right? Why this research?
Unfortunately, tuberculosis still occurs frequently—not very much in the Netherlands, but certainly
outside. The infection is also getting closer, not only due to travel and migration but also because of an
increase in difficult-to-treat cases, for example due to antibiotic resistance.
Every day, more than 4,000 people die from tuberculosis, and that is far too many. In the fight against
tuberculosis and other infectious diseases, we want to better understand how the immune system
responds before and after vaccination with the BCG vaccine. BCG has been used for over 100 years, but
we still have questions about the response to this vaccine.
The knowledge we gain from the BCG vaccination study is not specific to tuberculosis alone, but can also
be applied to improving vaccines against other infectious diseases. A win-win situation.
The BCG Vaccination Study
The BCG vaccination study is being conducted at the LUMC.
We want to vaccinate healthy volunteers (18-55 years old, men and women) with BCG. We know that
people can respond very differently to BCG vaccination, but we don’t know why these responses vary so
much.
What happens?
All participants will be vaccinated free of charge with:

The BCG vaccine on day 0

A single dose of rabies vaccine after 6 months
Before the BCG vaccination, a small skin biopsy will be taken; at every visit, blood will be drawn and a
swab of the oral cavity will be collected.
In the skin, we will examine which immune system cells are present and whether these differ from
person to person. In the blood, we will investigate the function of the immune system, both before and
after vaccination.
How much time does it take?
If you participate, the BCG vaccination study will take 7.5 months for you, during which the study team
will ask you to come to the LUMC seven times for vaccination and study visits.
The first study visit takes about half an hour, the second visit about one hour, and the other visits
approximately fifteen minutes each. A reimbursement for expenses is available.
When can you participate?

You can participate if:

You are between 18 and 55 years old,

You live in the Leiden area,

You are healthy,

You have not been vaccinated with BCG or Rabies before.
You also need to be willing to come in 7 times over a period of 7.5 months.
Which vaccines are used?
The vaccines used are called BCG (Danish strain 1331) and Rabipur (against rabies, Bavarian Nordic A/S).
Both vaccines will be administered intradermally (into the skin). This is the standard method for BCG,
and although it’s not the standard method for Rabipur, the LUMC has extensive experience with this
approach. Rabipur has been used for over 30 years and is proven to be safe and effective.
Please note: The rabies vaccination you receive in this study does not provide full protection against
rabies, as only a single dose is given. If you want protection against rabies, please contact the public
health service (GGD).
